Autism spectrum disorders show diverse underlying genetics, a variety of affected brain cell types and a broad ‘spectrum’ of clinical pathologies, which altogether pose a challenge to effective clinical intervention. Using human pluripotent stem cell and CRISPR technologies, I am modeling the loss of high-risk autism genes in human cortical neurons, astrocytes and microglia for transcriptional and functional stratification. I am also introducing vascular and mural cells in this system to establish 3-D microtissues that can faithfully model autism in a dish. Altogether, this work will enable us to elucidate cell- specific contributions to autism pathogenesis, reveal convergent underlying molecular pathways and help identify potential therapeutic targets.
